Key Helix-Stabilizing Dipeptide for Semaglutide Synthesis
The His-Aib sequence is a conserved functional motif in the Semaglutide backbone. The Aib (α-aminoisobutyric acid) residue, with its gem-dimethyl steric structure, stabilizes the peptide's α-helical conformation, which is essential for high-affinity GLP-1 receptor binding and long-acting glucose-lowering performance.
This product provides the complete preassembled His-Aib dipeptide, enabling one-step incorporation onto the resin during peptide synthesis. Compared with sequential coupling of His and Aib monomers, it eliminates one coupling reaction, significantly reduces truncated impurities, improves the overall yield of Semaglutide API, and is well suited for large-scale industrial production.
Single-Step Mild Fmoc Deprotection for a Simplified Cleavage Process
The molecule contains no tert-butyl, Pbf, or other side-chain protecting groups. Only the N-terminal Fmoc group requires mild removal with piperidine before peptide chain elongation. The final global cleavage process does not require prolonged exposure to strong TFA conditions, reducing the risk of peptide chain degradation and ensuring compatibility with standard Fmoc solid-phase peptide synthesis (SPPS) workflows.
No Pharmacological or Cosmetic Activity
This intermediate contains a bulky hydrophobic Fmoc fluorenyl protecting group and has a large molecular size, making it unable to penetrate cell membranes or the skin barrier. It exhibits no intrinsic biological activities such as glucose regulation, collagen stimulation, skin whitening, or soothing effects. Direct injection, oral administration, or topical application may leave residual aromatic fluorenyl impurities, potentially causing skin or mucosal irritation. Therefore, it is not intended for pharmaceutical formulations or cosmetic applications.
Universal Building Block for Long-Acting GLP-1 Peptides
In addition to Semaglutide, this intermediate is suitable for the synthesis of various Aib-modified long-acting glucose-lowering peptides and research peptides.
