Key Helix-Stabilizing Dipeptide for Semaglutide Synthesis
The His-Aib sequence is a highly conserved functional motif within the Semaglutide backbone. The Aib (α-aminoisobutyric acid) residue, with its gem-dimethyl steric structure, stabilizes the peptide's α-helical conformation, which is essential for high-affinity GLP-1 receptor binding and prolonged glucose-lowering activity.
This product is supplied as a TFA salt, providing excellent solubility in solid-phase peptide synthesis (SPPS) coupling solvents without precipitation, thereby enabling more efficient coupling than the corresponding free-base form and significantly reducing material loss during manufacturing. The fully preassembled His-Aib dipeptide can be incorporated into the resin in a single coupling step, eliminating one coupling reaction compared with sequential His and Aib incorporation. This substantially reduces truncated by-products and is ideally suited for large-scale industrial peptide production.
Single-Step Mild Fmoc Deprotection for Simplified Processing
The molecule contains no tert-butyl, Pbf, or other side-chain protecting groups. Only the N-terminal Fmoc group requires removal under mild piperidine treatment before further peptide elongation. Since no additional side-chain deprotection is required, the final cleavage process avoids prolonged exposure to concentrated TFA, minimizing peptide degradation and making the product fully compatible with standard Fmoc solid-phase peptide synthesis (SPPS) workflows.
No Pharmacological or Cosmetic Activity
This intermediate contains a bulky hydrophobic Fmoc fluorenyl protecting group and exists as a trifluoroacetate (TFA) salt. Due to its large molecular size, it cannot penetrate cell membranes or the skin barrier and exhibits no intrinsic biological activities such as glucose regulation, collagen stimulation, skin whitening, or soothing effects. Direct injection, oral administration, or topical application may leave residual fluorenyl or fluoride-related impurities, potentially causing skin or mucosal irritation. Therefore, it is not intended for pharmaceutical formulations or cosmetic applications.
Universal Salt-Form Building Block for Long-Acting GLP-1 Peptides
In addition to Semaglutide, this intermediate is suitable for the synthesis of various Aib-modified long-acting GLP-1 peptides and research peptide candidates. The TFA salt form significantly improves solubility compared with the corresponding free base, overcoming poor dissolution and incomplete coupling during peptide synthesis. It is a specialized preassembled building block for industrial production of long-acting GLP peptide therapeutics.
